Transgenic Breeding 桵y lab group has devised the first facile gene transfer method for the worldwide preeminent edible mushroom, Agaricus bisporus. Since traditional breeding approaches have not led to any notable improvements in commercial mushroom strains for more than 25 years, gene transfer technology now provides entirely new and exciting vistas for germplasm enhancement. Aside from improving agronomic traits, (e.g. disease resistance, quality, etc.), I am keenly interested in applying transgenic methods to the production of human biopharmaceuticals in mushrooms. Innate features of A. bisporus offer the potential of a high-capacity, economical, and secure system for drug manufacture. A mushroom-based biofactory for therapeutic proteins would provide commercial spawn makers with a new dimension of mushroom products having market values far exceeding that of the conventional food end usage.
Mushroom Pathologies 桝n aspect of my research program focuses on the biology, molecular biology, and diagnosis (PCR tests) of the major diseases (Verticillium, Trichoderma, and La France diseases) limiting commercial mushroom cultivation in North America. I am particularly interested in advancing our understanding of the viral complex associated with La France disease. Here, we have characterized three RNA viruses in A. bisporus, and are now exploring transgenic resistance.
Innovations for Mushroom Cultivation 桰 am collaborating with colleagues in the Penn State Department of Agricultural and Biological Engineering in the design of a complete mushroom production scheme for A. bisporus that utilizes grain-based substrates substituted for traditional manure-based compost. This technology addresses problems related to outdoor composting and compost disposal, and potentially offers a sterilized substrate for biopharmaceutical manufacture.
